Abstract

The invention relates to a method for detecting individual molecules. The method makes it possible to obtain an extremely good signal-to-noise ratio using non-linear optical effects. A sample support (5) is covered with a layer (4). Said layer has optically non-linear properties, and in particular is suitable for frequency doubling. A molecule of dye (2) is immobilised on the layer (4)and a laser beam (1) is focused onto the molecule through the sample support with limited diffraction. The wavelength of the laser beam corresponds to double the absorption wavelength of the dye molecule. The polarisation of the laser beam is selected according to the strong non-linear susceptibility coefficients of the second order of the optically non-linear layer. Wesentlich ist in jedem Fall, daß die Detektion kon­ fokal erfolgt, daß also der Photodetektor ein möglichst kleines Raumgebiet sieht, da das Hintergrundsignal (Rayleigh- und Ramanstreuung) proportional zum Volumen des vom Photodetektor gesehenen Raumgebietes ansteigt, während das optische Signal eines einzelnen Moleküls unverändert bleibt. Nachteilig bei diesem Verfahren ist sei­ ne beschränkte räumliche Auflösung, welche im wesentlichen durch die Ausdehnung des auf der Oberfläche gebildeten Laserfokus gegeben ist, der nicht kleiner als der Abbe-Limit der fokussierenden Optik sein kann. 1. Ausführungsbeispiel1st embodiment

Im Folgenden wird auf Fig. 1 Bezug genommen. In der einfachsten Ausführung der Vorrichtung werden die zu detektierenden Moleküle auf einer dünnen Schicht 4 eines optisch nichtlinearen Materials immobilisiert. Die Schicht selbst ist auf dem op­ tisch transparenten Substrat 5 (z. B. Mikroskopie-Deckgläschen) aufgebracht. Die Schichtdicke der Schicht 4 ist von der Größenordnung oder kleiner als die Wellenlän­ ge des anregenden Lichts. Das Material der Schicht 4 muß große Koeffizienten für Frequenzverdopplung bei der Frequenz des Anregungslichts besitzen.In the following, reference is made to FIG. 1. Oder man verwendet, bei senkrechtem Einfall, Laserlicht mit sogenannten Hermite-Gauss- Moden höherer Ordnung, bei welchen es im Laserfokus auch eine Feldkomponente mit starker Polarisation längs der Laserstrahlachse gibt [L. Novotny, E. J. Sanchez, X. S. Xie, "Near-field optical imaging using metal tips illuminated by higher-order Hermite-Gaussian beams", Ultramicroscopy 71(1-4), 21-9, 1998]. Coatings in which in fact only coefficients of the type χ izz with i ∈ {x, y, z} occur can in principle be used. Then the laser light must be irradiated at a non-perpendicular angle. Anstelle eines nichtlinearen Effektes zweiter Ordnung für Frequenzverdopplung kann auch ein nichtlinearer Effekt dritter Ordnung für eine Frequenzverdreifachung genutzt werden. Effekte dritter Ordnung kommen auch in isotropen Materialien vor. Das hätte den Vorteil, daß eine genaue Ausrichtung der Moleküle der Schicht 4 nicht mehr nötig ist. Auch eine genaue Ausrichtung der Polarisationsebene des Lichtstrahls 1 ist dann nicht nötig. Für die Beschichtung können konventionelle und einfache Techniken, wie z. B. spin coating, verwenden werden. Eine Vielzahl von als Material der Schicht 4 geeigneten Farbstoffen mit hohen Suszeptibilitäten dritter Ordnung fin­ det sich in dem Buch "Materials for Nonlinear Optics. Chemical Perspectives" von S. R. Marder, J. E. Sohn und G. D. Stucky, erschienen bei ACS, Washington, 1991.Instead of a second order nonlinear effect for frequency doubling, a third order nonlinear effect can also be used for frequency tripling. Als Schichtmaterial kommen z. B. Lithiumniobat, Kaliumniobat, Kaliumtitanyl­ phosphat oder organische Moleküle mit hohem Suszeptibilitätskoeffizienten in Frage. Als organische Moleküle eignen sich besonders eine Vielzahl von Farbstoffen unter­ schiedlicher Farbstoffklassen, z. B. Nitroaniline, Azobenzene oder Hemicyanine. Zwei geeignete kommerziell erhältliche Farbstoffe wären Disperse Red 1 und Stilbazolium. Die geeigneten Farbstoffe haben alle die in Fig. 4 gezeigten Grundstrukturen, wobei A einen Elektronenakzeptor und D einen Elektronendonor darstellen, welche über die gezeigte konjugierte Struktur miteinander verbunden sind. Die Zahl der in der Litera­ tur für Frequenzvervielfachung getesteten Stoffe ist enorm, und alle haben etwa die gleiche oben gezeigte Grundstruktur [s. z. B.: I. Ledoux, J. Zyss, "Polar and non-polar organic molecules, crystals and LB films for quadratic nonlinear optics", in: "Organic Molecules for Nonlinear Optics and Photonics", Eds.: J. Messier, F. Kajzar, P. Prasad, NATO ASI 194, Kluwer Academic, Dordrecht 1991].As layer material such. B. lithium niobate, potassium niobate, potassium titanyl phosphate or organic molecules with a high susceptibility coefficient in question. A large number of dyes with different classes of dyes are particularly suitable as organic molecules. B. nitroanilines, azobenzenes or hemicyanines. Die optische Anregung erfolgt durch einen Laserstrahl 1, welcher durch ein (nicht gezeigtes) herkömmliches Mikroskopobjektiv mit hoher numerischer Apertur beugungsbegrenzt durch das Substrat 5 in die Schicht 4 fokussiert wird. Der Laser­ strahl 1 wird bevorzugt von einem gepulsten Laser generiert, da bei einem gepulsten Laser die Konversionseffizienz bei der Frequenzvervielfachung wesentlich höher ist als bei einem kontinuierlichen Laser. Es kann zum Beispiel ein Titan-Saphir-Laser mit Femtosekundenpulskompression, welcher durch einen Argonionen-Laser gepumpt wird, zur Anwendung kommen. Durch den Energietransfercharakter der Anregung wer­ den außerdem nur Moleküle effizient angeregt, welche sich in einer Entfernung von wenigen bis etwa 100 Nanometern zur Oberfläche befinden (innerhalb des sogenann­ ten Försterradius). Da die Effizienz z. Das optische Signal wird durch einen (nicht gezeigten) herkömmlichen dichroitischen Spiegel vom Anregungslicht getrennt und über (nicht gezeigte) herkömmliche optische Filter, wel­ che Anregungslicht und frequenzvervielfachtes Licht gegen das erzeugte optische Si­ gnal diskriminieren, auf einen (nicht gezeigten) Einzelphotonen-empfindlichen De­ tektor gelenkt, der die detektierten Photonen in elektronisch weiter verwertbare Si­ gnale umwandelt. 2. Ausführungsbeispiel2nd embodiment

Im Folgenden wird auf Fig. 2 Bezug genommen. Der Aufbau ist analog zu dem­ jenigen gemäß Fig. 1, jedoch ist zwischen nichtlinearer Schicht 4 und Substrat 5 eine dielektrische optische Filterschicht 8 (analog zur optischen Schicht eines dichroiti­ schen Filters) eingebracht, welche einen hohen Reflexionskoeffizienten für die Anre­ gungswellenlänge hat, das in den unteren Halbraum abgestrahlte optische Signal aber weitestgehend durchläßt. Die optische Anregung erfolgt ebenfalls durch einen beu­ gungsbegrenzt fokussierten Laserstrahl 1, aber diesmal von oben. Die Detektion des erzeugten optischen Signals erfolgt entweder in Rückwärtsrichtung durch dasselbe Mikroskopobjektiv, durch das auch der Laserstrahl 1 fokussiert wurde, oder in Rich­ tung 6 durch ein zweites Mikroskopobjektiv.In the following, reference is made to FIG. 2. 3. Ausführungsbeispiel3rd embodiment

Bei diesem Ausführungsbei­ spiel erfolgt die optische Anregung nicht direkt durch einen senkrecht fokussierten Laserstrahl, sondern durch das lokalisierte elektromagnetische Feld am Spitzenapex einer metallischen Spitze 9. Bei dieser Spitze soll es sich um die Spitze eines her­ kömmlichen Atomkraftmikroskops handeln. Die Spitze bzw. Sonde 9 hat einen feld­ verstärkenden Effekt für das eingestrahlte Anregungslicht 11. Der Krümmungsradius der Spitze am Spitzenapex ist wesentlich kleiner als die Wellenlänge des Anregungs­ lichts 11. Vorzugsweise beträgt der Krümmungsradius wenige Nanometer.In the following, reference is made to FIG. 3. The excitation light 11 is laterally focused on the tip 9 , as z. B. in conventional optical near-field microscopes to detect a side vibration of the optical fiber there. Claims (11)

1. Verfahren zur optischen Detektion eines Partikels (2),
wobei ein Probenträger (5) mit einer Schicht (4) aus Material mit optisch nicht­ linearen Eigenschaften mit geeigneter Ausrichtung belegt wird;
wobei das Partikel (2) auf oder in der nichtlinearen Schicht (4) immobilisiert wird;
wobei ein Lichtstrahl (1) aus geeigneter Richtung mit geeigneter Polarisation auf das Partikel fokussiert wird, wodurch das fokussierte Licht in der nichtlinearen Schicht (4) teilweise frequenzvervielfacht (3) wird,
wobei die Wellenlänge bzw. Frequenz des Lichtstrahls (1) derart gewählt wird, daß das Partikel (2) bei der vervielfachten Frequenz optisch nachgewiesen werden kann;
wobei das Partikel (2) durch die frequenzvervielfachte Strahlung (3) angeregt wird; und
wobei das aufgrund der Anregung erzeugte optische Signal (7) detektiert wird.
1. Method for the optical detection of a particle ( 2 ),

2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß der Lichtstrahl (1) von der dem Partikel (2) abgewandten Seite des Probenträgers (5) auf das Partikel fokussiert wird.2. daß der Lichtstrahl (1) mit der genannten Wellenlänge von der beschichteten Seite des Probenträgers (5) aus auf das Partikel (2) fokussiert wird, wodurch das fo­ kussierte Licht an der reflektierenden Schicht (8) reflektiert und in der nichtlinearen Schicht (4) teilweise frequenzvervielfacht (3) wird.

daß eine metallische Spitze (9) an das Partikel (2) herangeführt wird;
daß der Lichtstrahl (11) derart auf das Partikel (2) fokussiert wird, daß sich eine von Null verschiedene Komponente des elektrischen Feldes des Lichtstrahls parallel zur metallischen Spitze (9) ergibt;
daß der Lichtstrahl (11) in der nichtlinearen Schicht (4) eine elektromagnetische Schwingung mit einem Vielfachen der genannten Frequenz anregt (3); und daß das Partikel (2) durch die elektromagnetische Schwingung (3) angeregt wird.

Verfahren nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, daß als Material für die nichtlineare Schicht (4) organische Moleküle gewählt werden, wobei die organischen Moleküle einen Elektronenakzeptor und einen Elektronendonor aufweisen, welche über eine elektronisch konjugierte Struktur miteinander verbunden sind.6. einer Fokussieroptik zum Fokussieren des Lichtstrahls (1) aus geeigneter Rich­ tung und mit geeigneter Polarisation auf das Partikel (2), wodurch ein optisches Signal erzeugt wird;
einer Sammeloptik (6) zum Sammeln des erzeugten optischen Signals; und mit
einer Detektionseinrichtung (1) zum Detektieren des erzeugten optischen Si­ gnals, wobei die Sammeloptik (6) das gesammelte optische Signal auf die Detek­ tionseinrichtung abbildet.
